TriSleep™ Playbook — A Compliant Evening Functional-Food Line

Playbook scope: a from-0-to-1 delivery plan for an evening-routine functional-food line, built for brand owners and channel operators. This playbook publishes methods and definitions only — no efficacy claims. Specs, MOQs, and lead times below are indicative until confirmed in the technical documentation and a formal quote.

Evening wind-down — quick-dissolve ritual

TriSleep™ is the ORAL sleep track within TONGNIAN's thirteen ™ product lines. Under the title "Oral Functional-Food Track from 0 to 1," Eight sections take the line from positioning to quoting. Every one stops at methods and definitions, never at performance promises.

01 · Market Positioning: The Evening-Routine Segment and the Format Opening

The problem this playbook solves: sleep-adjacent products force a choice between prescription weight and herbal vagueness — there is no compliant daily-ritual format carrying a new-material story with real documentation.

Sleep-adjacent products split into two camps. Prescription-grade aids carry dependency and compliance weight; herbal supplements are gentle but variable, and their ingredient stories are worn thin. Between the two sits an opening for a "daily-ritual" product: food-track compliant batch to batch, with a new-material narrative a channel can actually merchandise.

TriSleep™ fills that opening: fermented marine-derived EVs plus plant exosome fractions, pressed into a sublingual quick-dissolve tablet. Channel logic works backwards to format: cross-border e-commerce and evening-wellness gifting favour a 30-count jar (one month = one course = one gift box); pharmacy-adjacent retail favours blister singles for trial. Both tracks share one EV raw-material platform and one characterization baseline, so line extensions do not re-open development.

02 · Scientific Rationale: Oral EV Delivery and the Sublingual Choice

The design principle is cargo protection, not pharmacology. Exosomes are nanoscale vesicles that carry proteins, lipids, and microRNAs [1]; published human work on milk-derived exosomes showed that encapsulating a poorly bioavailable polyphenol (ellagic acid) inside exosomes measurably improved its oral bioavailability over a two-week intervention (Molecular Nutrition & Food Research, 2026) [3]. EVs act as delivery vehicles that shield cargo through the digestive environment — the reason an oral EV system is a credible functional-food chassis rather than a novelty.

Route is the second design variable. Sublingual absorption partially bypasses first-pass metabolism. Just as important for an evening program, the tablet removes the water-and-prep friction that erodes nightly adherence. The functional-food framing is deliberate: the product competes on format consistency and material documentation, while functional claims, where a market allows them, ride the local filing rather than the label's imagination.

Route comparison — sublingual vs swallowed

Fig. 1 — Route schematic: the sublingual format removes preparation friction and partially bypasses first-pass metabolism via buccal absorption.

03 · Formulation Design: The Dual-Source EV Family

Three formulation decisions define the track:

  1. Dual-source EV system. Fermented marine-derived EVs (scalable and standardizable at the fermentation upstream) co-formulated with plant exosome fractions — one characterization baseline across the whole family, so tablets, liquids, and stick packs are siblings, not separate projects.
  2. Quick-dissolve base. Sugar-free, neutral evening flavour; dissolves on the tongue in seconds. Adherence lives or dies on format — so the format is engineered first: it dissolves in seconds, no water, no prep.
  3. Format family on one baseline. Flagship: 30-count sublingual jar. Extensions: oral liquid / drops for liquid-preferring markets, single-serve lyophilized stick packs for tea-style rituals. Each format ships with a specification card (fill, packaging, stability plan — values per the technical agreement).

04 · Process: From Closed Micro-Batch to Finished Tablet

  1. EV material production — the fully enclosed 0.5–5 L micro-batch line: seed revival and expansion in closed tubing, scheduled harvest, tangential-flow separation and concentration (NuoqTFF 1.0 mother unit), sterile filtration; retain samples kept between batches. The downstream train layout follows published industrial guidance [4].
  2. Co-formulation — marine EVs blended with plant exosome fractions; payload and stability checked on retained samples against the characterization baseline.
  3. Finished-goods manufacture — tablet compression or liquid filling under food-grade GMP, with the EV baseline documentation attached to each production lot.
  4. Batch release — COA and batch records per batch; retain-sample plan as standard.

05 · QC & Characterization: Batch-Release Test Panel

Panel design follows MISEV2018 [2] and the national measurement guideline [5].

06 · Regulatory Pathway: US & EU Supplement Frameworks

The track is engineered for US and EU market entry first. In the US, the line is positioned as a dietary supplement under FDA's DSHEA framework — structure/function-style label discipline with the standard disclaimer, and ingredient documentation that stands up to FDA review. In the EU, oral formats route through the food-supplement framework, with novel-food assessment where ingredient history requires it. The material line holds FDA DMF Type II 044197 and INCI 41316 documentation — depth most supplement-shelf competitors cannot show, and leverage for premium pricing.

Claims discipline is written into the playbook: format and ritual language on pack ("quick-dissolve," "evening ritual," "30-day course"); functional claims only within what the destination-market framework authorizes (US structure/function rules, EU permitted-claims lists). The compliance team tracks FDA and EFSA movements and shares updates at playbook review points. For China market entry later, the blue-hat functional-food route is an additional option on the same base formula.

07 · Expected Program Outcomes: Framing & Evidence Level

  • Adherence is the design goal. A 30-count jar is a one-month course; the program metric is ritual consistency, not promises. The quick-dissolve format exists because consistency is the hard part of any evening program.
  • Perceived wind-down support is tracked in TONGNIAN's internal observational programs. Published oral-EV bioavailability work supports the delivery design [3]; it does not license therapeutic claims. Supporting observational evidence is available to partners: internal evening-program pilot cohorts, 2025–2026, under NDA [6].
  • No claims are made regarding sleep onset, duration, or any physiological outcome outside the authorized filing scope. Authority comes from documentation, batch measurement, and format honesty.
